Buserelin acetate-induced spermiation in Leptodactylus ocellatus and evaluation of semen characteristics - doi: 10.4025/actascibiolsci.v35i1.14777 Biological Sciences

By understanding the hormonal action behind the induction of spermiation in Leptodactylus ocellatus can help in the collection of semen for artificial insemination and cryopreservation. The experiments were conducted at the Experimental Frog Farm of the Federal University of Viçosa. Seven L. ocellatus males with the following secondary sexual characteristics were selected: black thorns, developed forelimbs, and amplexus reflex. Spermiation was induced in these animals by applying daily 0.4 μg buserelin acetate hormone (GnRHa) for 7 days, being the semen collected 90 min after the application. The volume, color, vigor, motility, sperm concentration, and spermatozoa morphology were subsequently evaluated. Three animals responded to GnRHa induction, enabling...

Gamete quality and management for in vitro fertilisation in meagre (Argyrosomus regius) ArchiMer
Ramos-júdez, Sandra; González, Wendy; Dutto, Gilbert; Mylonas, Constantinos C.; Fauvel, Christian; Duncan, Neil.
The aquaculture of meagre (Argyrosomus regius) requires methods for the control of reproduction that enable the production of families from specific individuals for selective breeding programs. We experimentally determined the parameters required for an in vitro fertilisation protocol. A total of 14 females and 5 males (mean ± S.D. weights of 20.45 ± 6.22 and 15.94 ± 2.75 kg, respectively) were used. Selected females had vitellogenic oocytes >550 μm in diameter and males had fluid sperm upon application of abdominal pressure. Both sexes were treated with an injection of 15 μg kg−1 of gonadotropin-releasing hormone agonist (GnRHa) to induce oocyte maturation/ovulation and enhance sperm production. To determine the timing of ovulation and window of high...

Slow-release GnRHa treatment prevented atresia during vitellogenesis and induced ovulation of captive wreckfish (Polyprion americanus) ArchiMer
Fauvel, Christian; Suquet, Marc; Severe, Armelle; Mylonas, Constantinos C.; Papandroulakis, Nikos.
The potential of wreckfish (Polyprion americanus) to breed in captivity was examined. Ovarian activity varied seasonally, with vitellogenesis beginning in November and ending in May. Slow-release implant with gonadotropin-releasing hormone agonist (GnRHa) prevented atresia, and both GnRHa injections or implants at the end of vitellogenesis triggered ovulation after 7 days. Spontaneous spawning was obtained only in one female, and the few eggs produced were of poor viability. Manual stripping and artificial fertilization produced viable gametes and embryos. Therefore, present captivity conditions inhibited final oocyte maturation (FOM) and spawning, but not vitellogenesis and spermatogenesis. GnRHa treatments stimulate FOM and ovulation of viable eggs.
